背景信息	The protein currently named Dihydrolipoyllysine-residue acetyltransferase component of pyruvate dehydrogenase complex, mitochondrial, or alternatively, 70 kDa mitochondrial autoantigen of primary biliary cirrhosis (PBC), Dihydrolipoamide acetyltransferase component of pyruvate dehydrogenase complex, M2 antigen complex 70 kDa subunit, or Pyruvate dehydrogenase complex component E2 (PDC-E2/PDCE2) and encoded by the human gene named DLAT or DLTA is part of the large multi-enzyme complex within the mitochondria that catalyzes the overall conversion of pyruvate to acetyl-CoA and carbon dioxide thus linking the glycolytic pathway to the tricarboxylic acid cycle. DLAT is the second enzyme in the chain and DLAT itself forms a huge multimeric complex of about 48 DLAT molecules within each massive pyruvate dehydrogenase complex. Found in the mitochondrial matrix and expressed in nearly all cells, defects in DLAT are the cause of primary biliary cirrhosis, a chronic progressive cholestatic liver disease; patients show autoantibodies to DLAT in their serum. Other defects in DLAT are the cause of Pyruvate dehydrogenase E2 deficiency which manifests as primary lactic acidosis and neurological dysfunction in infancy and early childhood.
