背景信息 GRB-2 (growth factor receptor-bound protein 2) is structurally and functionally similar to the C. elegans protein sem-5. GRB-2 contains one src homology 2 (SH2) domain and two SH3 domains that interact with EGFRs and PDGFRs and seems to be play an important part in the growth factor control of ras signaling. Ras signaling is involved in cell proliferation and differentiation and GRB-2 may be involved in the development of cancer cells. GRB-2 has also been found to have an impact on apoptosis. ISOFORM: GRB3-3 does not bind to phosphorylated epidermal growth factor receptor (EGFR) but inhibits EGF-induced transactivation of a RAS-responsive element. Isoform GRB3-3 acts as a dominant negative protein over GRB-2 and by suppressing proliferative signals, may trigger active programmed cell death.
