背景信息	The eukaryotic translation initiation factor 4B (eIF4B, eIF-4b) is one of several proteins involved in the initiation of eukaryotic translation. Together with other proteins EIF4B forms a complex with the 40S ribosomal subunit and Met-tRNA that ultimately allows for translation to begin. EIF4B contains two RNA binding domains, one binds RNA and the other specifically binds the 18S portion of the small (40S) ribosomal subunit RNA. EIF4B act as an anchor and then interacts with eIF4A, a helicase to help unwind and position the RNA for translation. EIF4B must be phosphorylated on Ser422 in order for translation to occur; the phosphorylation is mediated by S6K1/S6K2 kinases and or p90S6 kinase (RSK) in a growth factor dependent manner. Activation of S6K or RSK proteins requires PDK1 phosphorylation as well, thus two great signaling pathways, the mTOR/Pi3K and MAPK pathways converge on EIF4B to control its phosphorylation and subsequent activity. Phosphorylation of eIF4B at serine 406 is mitogen-regulated and is dependent on both MEK and mTOR activity.
