背景信息	MDA-5 or melanoma differentiation-associated protein 5 is a cytoplasmic protein with an interesting role. MDA-5 is one of a handful of proteins that have as part of their function to be natural immune receptors and sensors for viral nucleic acids. MDA-5 plays a major role in sensing viral infection and in the activation of a cascade of antiviral responses including the induction of type I interferons and proinflammatory cytokines. Its ligands include mRNA lacking 2'-O-methylation at their 5' cap and long-dsRNA (>1 kb in length). Upon ligand binding it associates with mitochondria antiviral signaling protein (MAVS/IPS1) which activates the IKK-related kinases: TBK1 and IKBKE which phosphorylate interferon regulatory factors: IRF3 and IRF7 which in turn activate transcription of antiviral immunological genes, including interferons (IFNs); IFN-alpha and IFN-beta. MDA-5 is the cellular sensor for detecting many virus types, including those from the Picornaviridae family such as Encephalomyocarditis virus and Mengo Encephalomyocarditis virus, as well as, West Nile, Dengue, Reo, and even dsDNA viruses like Vaccinia virus. Interestingly, some viruses have evolved mechanisms to turnoff or over-ride MDA-5’s sensory system. The Paramyxovirus was recently found to have a specific protein that directly interferes with the action of MDA-5 and thus prevent the anti-viral signaling that MDA-5 would normally initiate. Outside of innate viral detection MDA-5 also seems to function in assisting in NK cell function of suspicious cell types, growth inhibition and apoptosis. The protein is expressed at low levels in most cells and tissues with the highest expression seen in placenta, pancreas, and spleen and the lowest in brain, testis and lung. EMD-Millipore’s Anti-MDA-5 rabbit monoclonal antibody, clone EPR6743 has been tested in western blot on human THP-1 cell lysates.
