背景信息	FoxO, or FOX proteins (Fork head proteins) are transcription factors that control cell proliferation and survival. They function as tumor suppressors and are associated with longevity in several organisms. Fork head box protein O4, also known as Fork head domain transcription factor AFX1, with gene names FOXO4, AFX, AFX1 or MLLT7 is a transcription factor protein critical to the regulation of insulin signaling. FOXO4 activated insulin responsive genes by binding to the insulin responsive elements (IREs). FOXO4 also activates the transcription of insulin-like growth factor binding proteins, including IGFBP1. Interestingly, FOXO4 actively causes a decrease in expression hypoxia-induced transcription including down regulating the expression of their activating expression factor HIF1A. FOXO4 also influences cell cycle progression in a negative manner, and is involved in the up regulation of proteasome activity in developing embryonic stem cells (ESCs). Like many transcription factors FOXO4 can be found in the nucleus and cytoplasm. FOXO4 activity is regulated by numerous means but a major way is through phosphorylation. AKT-mediated phosphorylation of FOXO4 at multiple sites (Thr-32, Ser-187 and Ser-262) causes translocation out of the nucleus and a deactivation of the IRE genes; however phosphorylation of FOXO4 by other kinases such as JNK during oxidative stress causes FOXO4’s translocation into the nucleus and the activation of a whole different set of genes. FOXO4 is also post-translationally and can become acetylated or deacetylated as well as mono-ubiquitinated, which again facilitates nuclear localization, whereas de-ubiquitination facilitates transport into the cytoplasm. EMD-Millipore’s Anti-FOXO4 Phospho (pS262) specifically detects FOXO4 only when phosphorylated at serine 262 and this rabbit monoclonal has been tested in western blots FOXO4 transfected 293T cell lysates.
