背景信息	Interferon regulatory factor 5 (IRF-5) is a transcription factor protein responsible in part for host immunity against extracellular pathogens. IRF-5 regulates the induction of interferons IFNA and INFB upon virus infection. IRF-5 also up regulates various inflammatory cytokines upon viral infection as well. Activation is via TLR7 or TLR8, and results in phosphorylation of serine and threonine residues in the C-terminal autoinhibitory region; phosphorylation stimulates dimerization and transport into the nucleus where IRF-5 interacts with CBP/p300 coactivator to initiate transcription of its response genes. Interestingly, genetic polymorphisms mapping to the IRF-5 gene have been associated with susceptibility to inflammatory bowel disease type 14 (IBD14), systemic lupus erythematosus type 10 (SLEB10) and rheumatoid arthritis. Beyond interferons and viruses, IRF-5 also is critical in regulating several other cellular functions. The transcription factor IRF-5 exerts crucial functions in the regulation of DNA damage-induced apoptosis, death receptor signaling, and macrophage polarization. Recently, the COP9 signalosome (CSN), a general modulator of diverse cellular and developmental processes, associates constitutively with IRF-5 and promotes its protein stability as well, thereby pushing IRF-5 influence on cellular biology and physiology even further. EMD-Millipore’s Anti-IRF-5 rabbit monoclonal antibody, clone EPR6094 has been tested by western blot on U937, Ramos, and THP-1 human cell lysates and on permeabilized U937 by flow cytometry.
