背景信息 Glutamate is the predominant excitatory neurotransmitter in the CNS. It activates ionotropic and metabotropic glutamatergic receptors that mediate a wide range of neural processes including cognition and neural development. Glutamatergic signaling is terminated by the rapid removal of glutamate from the synaptic cleft. GLT-1 (EAAT2; Glutamate/aspartate transporter II) is a member of the family of high-affinity glutamate transporters involved in the reuptake of glutamate. GLT-1 is localized mainly on astrocytes of the forebrain, and transports glutamate in a sodium-dependent manner across an electrochemical gradient. Disruption of GLT-1 function results in accumulation of glutamate and overactivation of glutamatergic receptors, which may lead to neurotoxicity.
