背景信息 Protein regulated in development and DNA damage response 1 (REDD-1 or REDD1), also known as DNA damage-inducible transcript 4 protein, also known as HIF-1 responsive protein RTP801, and encoded by the gene DDIT4 or RTP801, is an inhibitor of mTORC1 and in so doing regulates cell growth, proliferation and survival in many cells systems. REDD1 inhibition of mTORC1 is mediated by a pathway that involves DDIT4/REDD1, AKT1, the TSC1-TSC2 complexes and the GTPase RHEB. REDD1 also plays an important role in responses to cellular energy levels and cellular stress, including responses to hypoxia and DNA damage. For instance, REDD1 regulates p53/TP53-mediated apoptosis in response to DNA damage via its effect on mTORC1 activity. REDD1’s role in the response to hypoxia depends on the cell type; for example it mediates mTORC1 inhibition in fibroblasts and thymocytes, but not in hepatocytes. REDD1 also plays a role in host-viral defense and it is required for mTORC1-mediated defense against viral protein synthesis and virus replication. Finally in neurons, REDD1 inhibits neuronal differentiation and neurite outgrowth mediated by NGF through its effects on mTORC activity and also mediates neuronal apoptotic cell death during development and stress.
