背景信息 Krüppel-like factors (KLFs) are zinc finger-containing transcription factors with regulatory functions in cell growth, proliferation, differentiation, and embryogenesis. Krüppel-like factor 4 (Klf4) has diverse functionss and acts as both an activator and repressor of transcription. The Klf protein has been shown to activate its own transcription throught its interaction with CACCC core sequences. High expression of Klf4 is observed in differentiating layers of epidermis and a loss of expression results in a break-down of barrier function including protection against pathogen invasion and dehydration. KLF4 is also associated with other biological functions such as proliferation, differentiation and carcinogenesis in gastrointestinal tissues. After DNA damage, Klf4 has been shown to induce DNA repair pathways. UniProt汇总 FUNCTION: Transcription factor which acts as both an activator and repressor. Binds the CACCC core sequence. Binds to multiple sites in the 5'-flanking region of its own gene and can activate its own transcription. Required for establishing the barrier function of the skin and for postnatal maturation and maintenance of the ocular surface. Involved in the differentiation of epithelial cells and may also function in skeletal and kidney development.
SUBUNIT STRUCTURE: Interaction with the C-terminal domain of MUC1 enhances suppression of TP53/p53 transcription (By similarity).
SUBCELLULAR LOCATION: Nucleus
TISSUE SPECIFICTY: Highest expression in the colon. Lower levels in testis, lung and small intestine.
INDUCTION: By interferon-gamma in Stat1-dependent manner.
DISRUPTION: phenotype Death shortly after birth due to loss of epidermal barrier function resulting from perturbation of late-stage epidermal differentiation structures including the cornified envelope. When selectively deleted in the surface ectoderm-derived structures of the eye, embryos develop normally and adults are viable and fertile but mutants display down-regulation of Krt12 and Aqp5 and multiple ocular defects including corneal epithelial fragility, stromal edema, defective lens and loss of conjunctival goblet cells.
