背景信息	Solute carrier family 22 member 3 (SLC22A3), also named Extraneuronal monoamine transporter (EMT), or Organic cation transporter 3 (OCT3), with gene names SLC22A3, EMTH, or OCT3 plays an important role in the disposition and function of cationic neurotoxins, neurotransmitters, and organic cations in the brain and other tissues. SLC22A3 does so by mediating the transport of these organic cations in and out of cells. It is a multi-pass membrane bound protein expressed chiefly in the brain and kidney but also in placenta, skeletal muscle, prostate, aorta, liver, fetal lung, salivary gland, adrenal gland but not in spleen. In the brain it is a major transporter for Histamine, Dopamine, Serotonin and Norepinephrine. Interestingly it is also the transporter for MPP+, a toxic chemical compound used in the neurotoxin MPTP and in the herbicide Cyperquat and Paraquat. These compounds compete with natural organic cations and reduce dopamine levels, inhibit the biosynthesis of catecholamines, deplete cardiac norepinephrine and inactivate tyrosine hydroxylase. These and other toxic effects lead to cessation of oxidative phosphorylation, ATP depletion, and cell death in neurons and parkinsonism like diseases in primates. Because of it link to dopamine transport SLC22A3 then, SLC22A3 is a highly studied target for research. In addition SLC22A3 also plays a critical role in the osmolarity regulation and fluid homeostasis of the brain and CNS. In the kidney and liver SLC22A3 is one a large family of polyspecific organic transporters that are critical for the elimination of many endogenous small organic cations as well as a wide array of drugs and environmental toxins. EMD-Millipore’s Anti-SLC22A3 rabbit monoclonal antibody has been tested in western blot on fetal human kidney, adult human muscle and placenta tissue lysates and in immunohistochemistry on paraffin embedded human kidney tissue sections.
