描述 FlowCellect™ Human Mesenchymal Stem Cell Characterization Kit 概述 Millipore’s FlowCellect™ Human Mesenchymal Stem Cell Characterization Kit provides a quantitative solution for clearly identifying human MSC cultures using flow cytometry. The kit contains four directly conjugated antibodies to phenotypically characterize hMSCs, with three positive expressing markers (CD105, CD90, and CD44), as well as one negative marker, CD19. These antibodies have been evaluated on a variety of different mesenchymal stem cell culture types such as adipocyte derived stem cells (ADSCs), bone marrow-derived hMSCs, and ES-derived hMSCs, ensuring that the kit can clearly monitor and characterize hMSC cultures from various types.
All kit antibodies are optimized for guava® bench top flow cytometers and provide a complete solution for hMSC analysis. However, this kit can be used on any flow cytometer following the same protocol, offering researchers a reliable, quantitative, and fully validated solution to study their MSC cultures. All four directly conjugated antibodies provided in the kit have been carefully evaluated to ensure optimal performance for flow cytometry, alleviating the need for any additional validation of the kit reagents. Also, plate-based stability testing and quality control was performed to confirm the stability of the antibodies over time. After 120 minutes (2 hours) post sample preparation, kit antibodies did not lose activity (< 5-10% of signal strength). 背景信息 Human mesenchymal stem cells are adult stem cells which have the capacity for multi-lineage differentiation, giving rise to a variety of mesenchymal phenotypes such as osteoblasts (bone), adipocytes (fat), and chondrocytes (cartilage). Human mesenchymal stem cells, or hMSCs, usually reside in the bone marrow stroma, but have also been indicated to be found in liver, spleen, peripheral blood, umbilical cord blood, and other mesenchymal tissues [2,4,6]. Due to their capacity for self-renewal over long periods of time and the ability to differentiate into specialized cells, interest in understanding the biology of MSC cultures has increased, especially for their potential use for therapy of a variety of diseases.
The International Society for Cellular Therapy (ISCT) has defined a list of guidelines which is used and widely accepted to appropriately identify hMSC cultures. The complexity and multi-lineage capacity of these cell types, can limit identification of a given cell type as MSC in nature. As outlined in the ISCT’s position statement, as well as reiterated by numerous resources (Pittenger et al., 1999, Docheva et al., 2008, and Chamberlain et al., 2007), the undifferentiated state of hMSC cultures can be phenotypically identified and characterized by a series of markers specific to hMSCs., specifically, MSC populations must be positive for at least several antigens, such as CD90, CD105, and CD73 [1,2,3,5,6]. Other resources also concluded that CD44, CD71, and Stro-1 are key markers which can be utilized to accurately validate hMSC cultures [2,3,5]. In fact, CD44 has been indicated to be an important receptor since it is involved in cell-matrix interactions which are shown to be expressed on MSCs. CD44 is abundantly found on chondrocyte cell surfaces and it is an important factor in maintaining cartilage homeostasis [3,7]. In addition to positive expression, negative markers have also been identified, producing less than 2% positive expression of these specific antigens.
