背景信息	CD32 is a cell surface receptor protein that is part of a large family of receptors localized on B-cells that act to modulate B-cell signaling. CD32’s other name is the low affinity immunoglobulin gamma Fc region receptor II-b. That means that it can bind IgG antibodies, when that happens on B-cells, it appears to cause a down regulation in IgG production by the B-cell. It is thought that this negative feedback loop by CD32 on B-cells helps regulate the level of IgG in the body. But CD32 is also expressed by most other immune cells, epithelial cells, and specialized cells such as the Langerhans cells. In fact, there are three subtypes of immunoglobulin gamma Fc-IgG receptors and CD32 is the most broadly distributed form. The receptors through binding of IgG (on macrophages for instance) help clear immune complexes and play a role in phagocytosis, however CD32's ability to bind IgG and activate signaling and immune responses also contributes to immune complex and auto antibody diseases such as vasculitis, rheumatoid arthritis, and autoimmune neutropenia. CD32 is also suspected in contributing to antibody dependent cell mediated cytotoxicity (ADCC), the system which mediates cell to cell interactions that lead to cell death and/or inflammation in normal and pathological autoimmune diseases. Thus CD32 is a target of intense research in hopes of using it to mediate our immune response, yet it cannot be blocked entirely as it is a key player in B cell antibody production as well. EMD-Millipore’s Anti-CD32 rabbit monoclonal antibody, clone EP6243 has been tested in western blot in both human cell line extracts and pancreatic tissue lysates, and in immunohistochemistry on paraffin embedded human spleen tissue samples.
