背景信息 Lysine-specific histone demethylase 1A (LSD1), (alternative names in older literature include BRAF35-HDAC complex protein BHC110, or Flavin-containing amine oxidase domain-containing protein 2) is encoded by the gene named KDM1A, AOF2, KDM1, KIAA0601 or LSD1 and is the histone demethylase that demethylates both 'Lys-4' (H3K4me) and 'Lys-9' (H3K9me) of histone H3, thereby acting as a coactivator or a corepressor, depending on the cellular context. LSD1 was one of the first lysine demethylases discovered and operates through a flavin-dependent (FAD) monoamine oxidase activity to remove histone methyl groups from Histone H3 complexes. During development LSD1 activity is essential for proper development beyond day 6.5 in mice and LSD1 plays a critical role in the expression and appropriate timing of key developmental regulators as part of the LSD1/CoREST/HDAC complex. Moreover, LSD1 interacts with numerous proteins through several different domains, most notably the tower domain, an extended helical structure that protrudes from the core of the protein that acts as the binding bridge between LSD1 and its targets. Defects in the domain can lead to severe developmental conditions, most notably in cardiac development, neuronal development and is also linked with several high-risk cancers. EMD-Millipore’s Anti-LSD1 rabbit monoclonal antibody has been tested in western blot against human, mouse and rat cell line lysates including lysates from human 293T, HeLa, Jurkat, PC-3 cell lines and rat C6, Raw 264.7 and PC-12 cell lines, as well as mouse NIH/3T3 cell lysates. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
