背景信息	The protein named DNA topoisomerase 1 and encoded by the human gene named Topoisomerase 1 is the enzyme that plays the essential role in unwinding DNA necessary for DNA replication and transcription. Topoisomerase 1 does so by inducing single-strand breaks via transesterification at the target site in duplex DNA. Topoisomerase 1 requires ATP for breakage of the ssDNA and is specifically inhibited by camptothecin, a plant alkaloid with anti-tumor activity. Expressed in all cells but highly expressed in rapid dividing or highly expressing cells such as endothelial cells and liver cells. In disease, auto-antibodies against Topoisomerase 1 are indicative of scleroderma autoimmune disease. Besides DNA Topoisomerase 1 also interacts with a variety of proteins necessary to perform its primary function including BTBD1, P53, Nucleolin, BTBD2, ASF/SF2, and UBE2I. Topoisomerase 1 is often up-regulated in cancers.
