背景信息	Receptor-interacting protein 1 (RIP, RIP1) or Cell death protein RIP, or more recently receptor-interacting serine/threonine-protein kinase 1 is a key serine-threonine kinase that transduces inflammatory and cell-death signals following activation of cell death receptors, activation of certain pathogen recognition receptors, oxidative stress, or DNA damage. RIP is involved in multiple cell death pathways including necrotic cell death and traditional apoptosis, as well as, cell survival signaling. More recently its role in a form of programmed necrosis or necroptosis has been found to be critical, and its inactivation by caspases cleavage essential to survival. In cell death signaling, the death domain of the RIP kinase binds to death receptors such as TNF receptor 1 (TNFR1), Fas, tumor necrosis factor-related apoptosis-inducing ligand receptor 1 (TRAILR1), and TRAILR2. It also associates with death domain-containing adaptor proteins, such as TNF-receptor-associated death domain (TRADD) and Fas-associated death domain (FADD) that are required for caspase-8 activation and apoptosis. Consistent with a role for RIP1 in regulating apoptosis, mice lacking RIP1 display extensive apoptosis and die at 1 to 3 days of age. Moreover, cultured cells lacking RIP1 are highly sensitive to TNF-induced cell death, possibly because they are unable to activate the prosurvival transcription factor NF-κB. Finally which path RIP follows appears to be determined by the level of ubiquitination. If RIP is ubiquitinated then it tends to play a role in cell survival, whereas if left un-ubiquitinated it plays its more traditional role in cell death, be it apoptosis or necroptosis. EMD-Millipore’s Anti-RIP rabbit monoclonal antibody has been tested in western blot against various human cell line lysates.
