背景信息	Neutral sphingomyelinase 1 (nSMase1) or more recently Sphingomyelin phosphodiesterase 2 is a multi-pass through membrane bound lipid phosphodiesterase that breaks sphingomyelin into phosphocholine and ceraminde. First identified in patients with a rare lysosomoal disease (Niemann-Pick disease), its pH optimum was near 7, thus neutral and hence the name, when compared to the other known sphingomyelinases, the acidic sphingomyelinases. The activation of nSMase1 has been suggested as a major route for the production of ceramide in response to cellular stresses. Such induction of ceramide production is thought to activate apoptosis in certain cellular situations. Neutral sphingomyelinase (nSMase1) appears to play a role in T-cell receptor (TCR) mediated apoptosis for instance, and specific induction of ceramide production only in the mitochondria specifically induced apoptosis in human MCF7 breast cancer cells. nSMase1 is membrane bound and has been localized throughout the intracellular membranes, notably in organelles such as the nucleus and mitochondria, and in the cytoplasm. EMD-Millipore’s Anti-nSMase1 rabbit monoclonal antibody has been tested in western blot on human cell line extracts and in fluorescent immunocytochemistry U87-MG cells in culture.
