描述 Anti-Axin-1 Antibody, clone 3F4.2 背景信息 Axin-1 is constitutively expressed and acts as a negative regulator of Wnt expression. It has been demonstrated that mouse Wnt, Wnt3a, is secreted by multiple cell lines and can act and as a growth factor for stem cells. Secreted Wnt3a is highly hydrophobic because it is palmitoylated on an essential cysteine C77; the palmitoylated state of Wnt3a is essential for stem cell signaling. Entrez基因汇总 This gene encodes a cytoplasmic protein which contains a regulation of G-protein signaling (RGS) domain and a dishevelled and axin (DIX) domain. The encoded protein interacts with adenomatosis polyposis coli, catenin (cadherin-associated protein), beta 1, 88kDa, glycogen synthase kinase 3 beta, protein phosphate 2, and itself. This protein functions as a negative regulator of the wingless-type MMTV integration site family, member 1 (WNT) signaling pathway and can induce apoptosis. The crystal structure of a portion of this protein, alone and in a complex with other proteins, has been resolved. Mutations in this gene have been associated with hepatocellular carcinoma, hepatoblastomas, ovarian endometriod adenocarcinomas, and medullablastomas. The interaction between axin and beta-catenin occurs via the armadillo repeats contained in beta-catenin. Ternary complex. Also binds to plakoglobin (gamma-catenin), APC, DVL and PP2A. Interacts with SMAD6, SMAD7 and RNF111. Interacts with DIXDC1; prevents interaction with MAP3K1. Interacts with MAP3K4, MDFI and MDFIC.

PTM: Probably phosphorylated by GSK3B and dephosphorylated by PP2A.
DISEASE: SwissProt: O15169 # Defects in AXIN1 are involved in hepatocellular carcinoma (HCC) [MIM:114550]. & Hypermethylation of the AXIN1 promoter may be associated with caudal duplication anomaly [MIM:607864]. Caudal duplication anomaly is characterized by the occurrence of duplications of different organs in the caudal region.
