应用说明 Western blotting at 1:200.
ELISA: 1:1000.
Immunoprecipitation: 1:100. Use rabbit capture secondary for best results.
Immunocytochemistry: light fixation with triton X-100 or acetone is recommended. 1:100.
Flow cytometry: 1:500 with permeabilization.
Immunohistochemistry: 1:100
RIA: 1:5000
Note: Catalog number MAB1622 is a more reactive and better antibody in most cases than MAB1685, particularly in non-human samples.
Optimal working dilutions must be determined by end user.
SUGGESTED WESTERN BLOT PROTOCOL FOR MAB1685:
1) Prepare protein samples by using appropriate lysis buffer e.g. RIPA (50 mM Tris HCl, 150 mM NaCl 5 mM EDTA, 1% Triton X-100, 100 mg/mL PMSF, 10 mg/mL leupeptin 20 mg/mL aprotinin).
2) Separate protein samples in SDS-PAGE 6% or 7.5%.
3) Blot onto the nitrocellulose membrane with TRANS-BLOT CELL (Bio-Rad) using constant current 100 mA overnight (Transfer buffer: 12.1 g Tris, 43.2 g glycine, 800 mL methanol to total volume of 4 liters).
4) Block membrane with 3% BSA in TBS (2.42 Tris, 8 g NaCl to total volume 1 liter, pH 7.6) 1-2 hours at room temperature.
5) Incubate membrane in diluted primary antibody solution (diluted in TTBS: 0.1% Tween-20 in TBS) one hour.
6) Wash membrane with TTBS: two quick rinses, 1 x 15 min, 3 x 5 min.
7) Incubate membrane with diluted HRP-conjugated secondary antibody solution (TTBS) for one hour.
8) Wash membrane with TTBS: 2 quick rinses, 1 x 15 min, 4 x 5 min.
