背景信息 Nuclear DFB-2-related (NDR) kinases are a highly conserved, subclass of the AGC protein kinase group that are known to be regulated by Mps One Binding (MOB) binding partner through binding and subsequent activation. They can also be activated by phosphorylation to the hydrophobic motif at the C-terminal and the kinase domain activation loop. NDR kinases play an integral role as regulators in several cellular processes such as; cytokinesis, mitotic exit, centrosome duplication, cell growth, morphogenesis, and development. NDR1 and NDR2 are two serine-threonine kinases belonging to the NDR kinase family that share significant homology with each other. NDR1 autophosphorylation is induced by Ca2+ in addition to phosphorylation by an unknown upstream kinase. Both NDR1 and NDR2 are nearly ubiquitous in expression, with the exception of NDR1 not being observed in the brain. In contrast, NDR2 has highest expression in the thymus. UniProt汇总 FUNCTION: Negative regulator of MAP3K1/2 signaling. Converts MAP3K2 from its phosphorylated form to its nonphosphorylated form and inhibits autophosphorylation of MAP3K2.
CATALYTIC ACTIVITY: ATP + a protein = ADP + a phosphoprotein.
COFACTOR: Magnesium.
ENZYME REGULATION: Activated by binding of S100B which releases autoinhibitory N-lobe interactions, enabling ATP to bind and the autophosphorylation of Ser-281. Thr-444 then undergoes calcium-dependent phosphorylation by an upstream kinase. Interactions between phosphorylated Thr-444 and the N-lobe promote additional structural changes that complete the activation of the kinase. Autoinhibition is also released by the binding of MOB1/MOBKL1A and MOB2/HCCA2 to the N-terminal of STK38.
SUBUNIT STRUCTURE: Homodimeric S100B binds two molecules of STK38. Component of the MLL5-L complex, at least composed of MLL5, STK38, PPP1CA, PPP1CB, PPP1CC, HCFC1, ACTB and OGT. Interacts with MOB1 and MOB2. Interacts with MAP3K1 and MAP3K2 (via the kinase catalytic domain).
