背景信息	Transforming protein RhoA (RHOA, ARH12, ARHA, RHO12) is part of a larger superfamily of proteins know as the Ras superfamily; they are all GTP binding proteins and each affects cell division, timing and regulation of cell growth and proliferation in some way. RHOA is known to regulate actin cytoskeleton interactions that link plasma membrane receptors to the assembly of focal adhesions and actin stress fibers and plays key roles in cell motility and cell division. RHOA operates through a key kinase called ROCK1 (Rho-associated, coiled-coil containing protein kinase 1). Also various receptors interact with RHOA; NgR1, LINGO1, neurotrophin receptor p75, TROY and many others all activate RHOA when bound by their respective ligands, which then in turn activates various ROCK1 facilitated signal pathways. In various tissues RHOA plays key roles in actin-facilitated growth extensions or retractions. For instance cell division RHOA is required for the myosin contractile ring formation and the correct operation during cytokinesis, and with keratinocytes RHOA is vital to proper apical cell junction formation, but in neurons RHOA activation leads to nerve growth cone collapse and the inhibition of the growth and repair of neural pathways and axons. RHOA is localized in the cell membrane on the cytoplasmic side but can translocate to the cytoplasm during cell division cycles. In cancer RHOA plays a key role in cancer metastasis by facilitating cell migration as well as division, and interestingly RHOA’s actions can be differentiated from brother proteins RHOB and RHOC. EMD-Millipore’s Anti-RHOA polyclonal antibody has been tested in western blot on human brain lysate and in paraffin embedded immunohistochemistry on human colon cancer tissue sections.
