背景信息	The protein named pyruvate carboxylase (PC), mitochondrial, or Pyruvic carboxylase (PCB) and encoded by the human gene named PC is the enzyme that catalyzes a 2-step reaction, involving the ATP-dependent carboxylation of the covalently attached biotin in the first step and the transfer of the carboxyl group to pyruvate in the second reaction. Pyruvate Carboxylase catalyzes in a tissue specific manner. The initial reactions are primarily done in the liver and kidney, while second (lipid synthesis from pyruvate) being more concerned with lipid metabolism are more focused in adipose tissue, liver, and brain. PC is an essential part of the Krebs Cycle in mitochondria and Pyruvate Carboxylase plays a crucial role in gluconeogenesis, lipogenesis, in the biosynthesis of neurotransmitters, and in glucose-induced insulin secretion by pancreatic islets. Oxaloacetate produced by Pyruvate Carboxylase is an important intermediate, which is used in many biosynthetic pathways as well. Because PC serves in a few important metabolic crossroads between lipid and carbohydrate metabolism, Pyruvate Carboxylase expression is highly regulated and responsive to metabolic conditions. Therefore, Pyruvate Carboxylase is a target for research in metabolic syndrome and diabetes. Defects in Pyruvate Carboxylase can cause lactic acidosis and hypoglycemia.
