背景信息	The protein Di-N-acetylchitobiase, encoded by the human gene named CTBS or CTB, is a lysosomal glycosidase involved in the degradation of asparagine-linked glycoproteins. These asparagines-linked glycoproteins are the basis of chitin, a complex polysaccharide that forms the structural basis for insect exoskeletons and fungal and plant cell walls. Though mammals express CTBS and its location in the lysosome infers that the enzyme may be expressed as a defensive mechanism rather than as a digestive or nutrient mechanism. Interestingly, human expression is quite strong when compared to that of other higher mammals such as pig and bovine, yet its precise function in human metabolism is not known. It is thought that deficiencies in CTBS activity could lead to a similar phenotype as that seen in knockout mice where investigators observed multilamellar inclusion bodies reminiscent of polar lipids present in epithelia of a scattered subset of proximal tubules in the kidney, and less constantly, enlarged Kupffer cells were observed in liver, filled with phagocytic material resembling partly digested red blood cells. Recent evidence shows that such deficiencies could be the cause of a not yet identified lysosomal storage disease. EMD-Millipore’s Anti-CTBS rabbit polyclonal antibody has been tested in western blot on MCF7 cell lysates, in paraffin embedded immunohistochemistry on human prostate cancer tissue sections and in fluorescent immunocytochemistry on HepG2 cells in culture.
