背景信息	The protein named NEDD4-like E3 ubiquitin-protein ligase WWP2 or Atrophin-1-interacting protein 2 (AIP2), or WW domain-containing protein 2 and encoded by the human gene named WWP2 is an important developmental ubiquitin E3 ligase. The ubiquitin proteasome degradation system is driven through a series of enzymatic reactions which ultimately result in ubiquitin being ligated to target proteins and subsequently degraded. The final ligation of ubiquitin to the target protein is accomplished via what are called “E3 ligases” and WWP2 happens to be the E3 ligase responsible for ligating ubiquitin on to the important developmental transcription factors POU5F1 and OCT4 as well as other proteins like EGR2, RPB1, SCNN1A, SCNN1B, SCNN1G, WBP1, WBP2, ATN1 and SLC11A2. Through ubiquitination and degradation the levels of many of these important proteins is maintained. WWP2 is expressed in most tissues and cell types with detectable expression observed in heart, throughout the brain, placenta, lung, liver, muscle, kidney, and pancreas. WWP2 is also highly expressed in pluripotent ES stem cells but expression is reduced after embryoid body formation though still detectable. Typically WWP2 is localized in the cytoplasmic arenas but can be found in the nucleus in ES cells. Interestingly WWP2’s own level is maintained through ubiquitination as well, as WWP2 autoubiquitinates itself through interaction with SCF(FBXL15) complex. EMD-Millipore’s Anti-WWP2 rabbit polyclonal antibody has been tested in western blot on mouse liver extracts and in paraffin embedded immunohistochemistry on human pancreas cancer tissue sections.
