背景信息 PEA-15 is a 15 kDa cytoplasmic protein which is abundantly expressed in astrocytes, although it has been detected in a variety of other cells including those of the heart and muscle. It may be phosphorylated on Ser104 by protein kinase C and on Ser116 by CAMKII, in a calcium-dependent manner, in response to various hormones and neurotransmitters. PEA-15 also contains an N-terminal death effector domain (DED) which is characteristic of proteins that regulate apoptosis. Previous studies have reported that it inhibits apoptosis induced by the signaling of TNFα in astrocytes. However, it is becoming increasingly clear that PEA-15 is a multifunctional protein affecting diverse cellular processes, possibly by its modulation of the MAP kinase signaling pathway. Previous studies have demonstrated that PEA-15 binds to ERK kinase in the cytoplasm, and prevents the activation and nuclear translocation of ERK.
