描述 Anti-Pramel-5 Antibody 背景信息 Pramel-5 was identified by Bortvin et al. (2003) as a gene whose expression pattern was like that of Octamer-4 (Oct-4), a gene with an essential role in early development. Criteria used for selection included: (1) expression in cleavage-stage embryos, blastocysts and in the embryonic germline, and (2) the genes should not be expressed in somatic tissues. Subsequent analysis showed that Pramel-5 was expressed in undifferentiated ES cells, normal 4-cell embryo, morula and blastocysts. No expression was detected in a panel of somatic tissues including adult brain, embryonic brain, embryonic limbs, adult skeletal muscle, lung, liver, spleen, kidney and heart. A comparison of the sequence of Pramel-5 with known genes showed that Pramel-5 is similar to the PRAME antigen detected in a wide range of human tumors (van Baren et al, 1998).
In addition to serving as a marker of pluripotency, Pramel-5 may also influence the success of nuclear cloning. Analysis of the expression of a number of Oct-4 related genes, including Pramel-5, in somatic and ES cell derived clones revealed that successful reactivation of these genes correlated with the development of cloned embryos to term. Blastocysts derived from nuclei of ES cells, where the genes are already active, retain their expression pattern as observed during embryogenesis. In contrast, 40% of the somatic derived blastocysts failed to reactivate these genes.
