Apaf1 binds to cytochrome C (Apaf2) and caspase-9 (Apaf3), which leads to caspase-9 activation. Activated caspase-9 in turn cleaves and activates caspase-3 which is one of the key proteases, being responsible for the proteolytic cleavage of many key proteins in apoptosis (Li et al. 1997). Apaf1 can also associate with caspase-4 and caspase-8 (Hu et al. 1998). This protein contains several copies of the WD-40 domain, a caspase recruitment domain (CARD), and an ATPase domain (NB-ARC). Upon binding cytochrome c and dATP, this protein forms an oligomeric apoptosome. The apoptosome binds and cleaves caspase 9 preproprotein, releasing its mature, activated form. Activated caspase 9 stimulates the subsequent caspase cascade that commits the cell to apoptosis.
