背景信息	PDGF, which is a human platelet derived growth hormone, is a tyrosine kinase receptor. PDGF is involved in cell proliferation, migration, and survival. There are two isoforms of PDGF, PDGF receptor-α and PDGF receptor-β. PDGF receptor-β can only bind two specific PDGF isoforms, PDGF-β and PDGF-Δ. Tyr751 and Tyr740 are required for PDGFR-β mediated PI3 kinase activation.

The identity of the growth factor bound to a receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, composed of both platelet-derived growth factor receptor alpha and beta polypeptides. This gene is flanked on chromosome 5 by the genes for granulocyte-macrophage colony-stimulating factor and macrophage-colony stimulating factor receptor; all three genes may be implicated in the 5-q syndrome. A translocation between chromosomes 5 and 12, that fuses this gene to that of the translocation, ETV6, leukemia gene, results in chronic myeloproliferative disorder with eosinophilia.

It is released by platelets upon wounding and plays an important role in stimulating adjacent cells to grow and thereby heals the wound. SUBUNIT STRUCTURE: Antiparallel disulfide-linked dimer of non-identical (A and B) chains. Homodimers of A and B chains are implicated in transformation processes. Interacts with XLKD1 By similarity. SUBCELLULAR LOCATION: Secreted. TISSUE SPECIFICITY: Expressed at high levels in the heart, brain (sustantia nigra), placenta and fetal kidney. Expressed at moderate levels in the brain (hippocampus), skeletal muscle, kidney and lung. INVOLVEMENT IN DISEASE: A chromosomal aberration involving PDGFB is a cause of dermatofibrosarcoma protuberans (DFSP) [MIM:607907]. Translocation t(17;22)(q22;q13) with COLA1. DFSP is an uncommon, locally aggressive, but rarely metastasizing tumor of the deep dermis and subcutaneous tissue. It typically occurs during early or middle adult life and is most frequently located on the trunk and proximal extremities. Pharmaceutical use Available under the name Regranex (Ortho-McNeil). Used to promote healing in diabetic neuropathic foot ulcers. MISCELLANEOUS: A-A and B-B, as well as A-B, dimers can bind to the PDGF receptor. SEQUENCE SIMILARITIES: Belongs to the PDGF/VEGF growth factor family.
