    背景信息 The protein named myosin light chain kinase from smooth muscle, or MLCK (smMLCK), or Kinase related protein (KRP), or Telokin and encoded by the human gene MYLK/MLCK/MLCK1/MYLK1 is the important kinase in smooth muscle cell contraction. MLCK is a calcium/calmodulin dependent myosin light chain kinase that phosphorylates the myosin light chains of smooth muscle. The phosphorylation will enable the myosin cross bridge to bind to the actin filament and allow contraction to begin; because smooth muscle does not contain a troponin complex as striated muscle does, this phosphorylated driven mechanism is the main pathway for regulating smooth muscle contraction. MLCK plays critical roles in cell survival, apoptosis, cell motility, wound healing, vascular injury repair, cellular tight junction transport and a host of other critical functions. MLCK is expressed in smooth muscle as well as in a variety of non-muscle cells in both adults and fetal tissues. It is localized in the cytoplasm and in various cellular projections depending upon the cell type and context being examined. Interestingly defects in MLCK are associated with Familial aortic aneurysm syndrome, a disease characterized by permanent dilation of the thoracic aorta usually due to degenerative changes in the aortic wall.
