背景信息 Cytohesin-1 is a guanine nucleotide exchange factor and an integrin binding protein belonging to a closely related group of proteins known as the cytohesion family. Cytohesin-1 contains 2 domains; a C-terminal, pleckstrin homology (PH) domain, and a central, large Sec7 domain. The PH domain serves to mediate the proteins functional regulation and the reversible membrane recruitment. The Sec7 domain acts as the primary regulator for phosphoinositide-dependent membrane attachment of the protein. Cytohesin-1 appears to have an integral role in integrin alphaLbeta2 adhesive function, and in the activation of integrin mediated pathways involved in wound healing, and immune system cell-cell/ cell-matrix interactions. Entrez基因汇总 The protein encoded by this gene is a member of the PSCD family. Members of this family have identical structural organization that consists of an N-terminal coiled-coil motif, a central Sec7 domain, and a C-terminal pleckstrin homology (PH) domain. The coiled-coil motif is involved in homodimerization, the Sec7 domain contains guanine-nucleotide exchange protein (GEP) activity, and the PH domain interacts with phospholipids and is responsible for association of PSCDs with membranes. Members of this family appear to mediate the regulation of protein sorting and membrane trafficking. This gene is highly expressed in natural killer and peripheral T cells, and regulates the adhesiveness of integrins at the plasma membrane of lymphocytes.
