背景信息 Paired box protein Pax-5, (PAX5) or B-cell-specific transcription factor (BSAP), is an important transcription factor in the maturation B-cells as well in the regulation of CD19, another B cell specific marker. PAX5 also appears to play a similar role in neural development and spermatogenesis. Its expression is developmentally regulated being expressed primarily only during early development and limited to the nucleus. As with many transcription factors, PAX5 has multiple isoforms at the protein level but they appear all function similarly. As a member of the paired box (PAX) family of transcription factors, it contains the central feature of this gene family, the unique and highly conserved DNA-binding motif known as the paired box. The PAX proteins are important regulators in early development, and alterations in the expression of their genes play important roles in neoplastic transformation and cancer. Mutations and abnormal expression of PAX5 have been associated in many cancers as well unusual diseases. For instance, the Reed-Sternberg cells of Hodgkin’s lymphoma (a classic cell type identified in the disease) almost always express Pax-5, and a chromosomal aberration involving PAX5 is a cause of acute lymphoblastic leukemia. EMD-Millipore’s mouse anti-PAX5 monoclonal, clone 7D3/1A7 has been tested in western blot against recombinant human PAX-5 protein as well as Raji lymphoblastoid cell lysates and human umbilical vein endothelial cell line EVC-304 cell lysates.
