    背景信息 Tumor Necrosis Factor α (TNF-alpha, cachectin) is a cytokine, produced by variety of cell types, that is involved in mediation of the inflammatory response, resistance to infection, and resistance to tumor growth. TNF-alpha is known to bind to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-α is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia. Studies have shown that under certain conditions it can stimulate cell proliferation and induce cell differentiation. The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells. Mutations in TNF-alpha have been associated with Psoriatic Arthritis (PSORAS): an inflammatory, seronegative arthritis associated with psoriasis. Human and mouse TNF-α are approximately 79% homologous at the amino acid level and exhibit cross-species reactivity. TNF-alpha induced protein 6 may be involved in cell-cell and cell-matrix interactions during normal inflammatory responses and during tumorigenesis.
