背景信息	Kininogen-1, otherwise known as Fitzgerald factor or the high molecular weight kininogen (HMWK), is the original precursor to several important thiol proteases involved in blood coagulation or circulation function. Kininogen-1 gets cleaved into six different products: Kininogen-1 heavy chain, T-kinin, bradykinin, lysyl-bradykinin, kininogen-1 light chain (LMWK) and low molecular weight growth promoting factor. The most interesting of the by-products of Kininogen-1 processing is bradykinin. It is produced from Kininogen-1 through proteolytic processing by the plasma protease kallikrein. Bradykinin has a number of effects depending upon the tissue from smooth muscle contraction, to induction of hypotension, mediation of inflammation to increasing vascular permeability and cardioprotective effects. High-molecular-weight kininogen (HMWK) is essential for blood coagulation and assembly of the kallikrein-kinin system. Through the action of various serum proteases (e.g. kallikrein, trypsin, plasmin) both HMWK and LMWK can generate bradykinin but only HMWK suffices for activating the factors need in coagulation. The protein is found in plasma and produced in the liver; T-kinin is detected in malignant cancers such as breast, colon and ovarian. EMD-Millipore’s Anti-Kininogen-1 rabbit monoclonal antibody, clone EPR6097 has been tested by western blot on human plasma, platelet, and ovarian cancer lysates and by immunohistochemistry on paraffin embedded human kidney tissue samples.
