Notes:
Slides/Fixation
cells should be suspended with PBS, pH 7.8 in 200 mM NaCl
air dried onto slides at room temp.
immediately fixed in -20ºC Methanol:Acetone;3:7 (v/v)
air dry slides and store @ 4ºC (<1 month) or -20ºC (6-12 months)
Staining
3 step amplification procedure must be used, i.e.
Mab + goat Anti-mouse IgG + Rabbit Anti-goat/FITC, or
Mab + goat Anti-mouse/biotin + Streptavidin/FITC, or
Mab + guinea pig complement + goat Anti-guinea pig C3/FITC
each incubation should be 45' at 37ºC
Comments
A 2-step staining procedure is not sensitive enough to detect the EBNA.
