背景信息 Adipose tissue is highly specialized to play important roles in energy storage, fatty acid metabolism, and glucose homeostasis. Excess of adipose tissue in obesity and lack of adipose tissue in lipodystrophic states are associated with insulin resistance and type-2-diabetes in human and animal studies. Several groups have reported the identification of mouse and human transcript encoding an adipocyte-specific secretory protein originally termed Acrp30 (other names include AdipoQ, apM1, GBP28 and adiponectin). Adiponectin is a protein of 247 amino acids consisting of an N-terminal collagen-like domain and a C-terminal globular domain. The globular domain shares significant homology to the globular domains of type VIII and X collagens, subunits of the complement factor C1q as well as hib27, the latter a protein found in the serum of hibernating mice during non-hibernation. The crystal structures for the globular domain of adiponectin at 2.1 Å resolution reveals significant homology to TNF (tumor necrosis factor) family. Adiponectin circulates as a homotimer or larger complexes of 12-18 subunits. The concentration in serum is approximately 5-10 μg/mL, which corresponds to a maximum concentration of 10-30 nM for the higher order complex.
Serum levels of adiponectin are significantly decreased in both obesity and type-2-diabetes, indicating a possible metabolic role or relationship to insulin resistance. In consistence with this, a recent report emphasized a proteolytic fragment (globular domain) of adiponectin to stimulate fatty acid oxidation by muscle.产品信息 格式 Ascites 演示 Ascites fluid. 应用说明 Western blot: 1:5,000-1:10,000 using ECL. Reacts with the 30 kDa adiponectin protein. Suggested blocking and dilution buffer is TBST with 0.05% Tween 20 with 5% skim milk. Suggested incubation time is 1 hour at room temperature. Human plasma is recommended as a positive control.
Immunoprecipitation: 1-10 μg in a final reaction volume of 700 μL. Final total protein concentration in the reaction mix: 1.6 mg/mL. Suggested capture agent is Protein A.
ELISA, direct and indirect: 1:5,000-1:10,000.

SUBUNIT: Homomultimer. Forms trimers, hexamers and 12- to 18-mers. The trimers (low molecular weight complexes / LMW) are assembled via non-covalent interactions of the collagen-like domains in a triple helix and hydrophobic interactions within the globular C1q domain. Several trimers can associate to form disulfide-linked hexamers (middle molecular weight complexes / MMW) and larger complexes (higher molecular weight / HMW). The HMW-complex assembly may rely aditionnally on lysine hydroxylation and glycosylation. LMW, MMW and HMW complexes bind to HBEGF, MMW and HMW complexes bind to PDGFB, and HMW complex binds to FGF2.

PTM: Hydroxylated Lys-33 was not identified in PubMed:16497731, probably due to poor representation of the N-terminal peptide in mass fingerprinting. & HMW complexes are more extensively glycosylated than smaller oligomers. Hydroxylation and glycosylation of the lysine residues within the collagene-like domain of adiponectin seem to be critically involved in regulating the formation and/or secretion of HMW complexes and consequently contribute to the insulin- sensitizing activity of adiponectin in hepatocytes (By similarity). & O-linked glycans consist of Glc-Gal disaccharides bound to the oxygen atom of post-translationally added hydroxyl groups. & Not N-glycosylated.
DISEASE: SwissProt: Q15848 # Defects in ADIPOQ are the cause of adiponectin deficiency [MIM:605441]. The result is a very low concentration of plasma adiponectin. Decreased adiponectin plasma levels are associated with obesity insulin resistance, and diabetes type 2.

MISCELLANEOUS: Variants Arg-84 and Ser-90 show impaired formation of HMW complexes whereas variants Cys-112 and Thr-164 show impaired secretion of adiponectin in any form. & HMW-complex blood contents are higher in females than in males, are increased in males by castration and decreased again upon subsequent testosterone treatment, which blocks HMW- complex secretion (By similarity).
