背景信息 A member of the Endophilin family, Endophilin A1 is a protein containing SH3 domains, and has been implicated in the trafficking of clathrin-mediated synaptic vesicle endocytosis within the membrane. It was originally characterized as a presynaptically enriched protein that binds to endocytic proteins polyphosphoinositide synaptojanin and dynamin. Endophilin A1 has been implicated in endocytic vesicle budding when its function in cell-free systems and in living synapses has been disturbed. Along with the SH3 domain, Endophilin A1 contains a BAR domain in the N-terminal region, that has been found to induce and bind membrane curvature. This protein’s expression is localized to the brain, most specifically in the frontal cortex. DOMAIN: An N-terminal amphipathic helix, the BAR domain and a second amphipathic helix inserted into helix 1 of the BAR domain (N-BAR domain) induce membrane curvature and bind curved membranes by similarity. The BAR domain dimer forms a rigid crescent shaped bundle of helices with the pair of second amphipathic helices protruding towards the membrane-binding surface.
