背景信息	The protein named Natural resistance-associated macrophage protein 2 (NRAMP 2), or Divalent cation transporter 1, or Divalent metal transporter 1 (DMT-1) and encoded by the human gene named SLC11A2/DCT1/DMT1/NRAMP2 is an important plasma membrane bound metal ion transport protein. Interestingly it was original cloned as a gene important for macrophage activation against intracellular pathogens, presumably because it allowed toxic metal ion transport. Today SLC11A2 is recognized as being particularly important for the transport of iron but SLC11A2 also transports manganese, cobalt, cadmium, nickel, vanadium and lead. SLC11A2 Is expressed by most cells but particularly in erythroid precursor cells and in neurons and glial cells. Tissue expression is observed in the brain, spleen, thymus, pancreas, small intestine and kidney. In neurons SLC11A2 forms a complex with NDFIP1 and NEDD4L upon iron and colbalt exposure and results in ubiquitination and ultimate destruction of SLC11A2 as a form of regulation, and in so doing helps prevent metal toxicity in human neurons. Defects in SLC11A2 lead to various metal toxicity diseases or with hypochromic microcytic anemia where the red blood cells (RBCs) very small because of malformed hemoglobulin.
