背景信息	Pyruvate dehydrogenase protein X component (PDHX) or alternatively dihydrolipoamide dehydrogenase-binding protein of pyruvate dehydrogenase complex, E3-binding protein (E3BP), or lipoyl-containing pyruvate dehydrogenase complex component X, or proX is an anchoring protein encoded by the human gene PDHX/PDX1. PDHX is required for anchoring of the dihydrolipoamide dehydrogenase to the dihydrolipoamide transacetylase core of the pyruvate dehydrogenase complexes in eukaryotic mitochondria. The anchoring is essential for proper functioning of the PDH complex which catalyzes the conversion of pyruvate to acetyl coenzyme A, thereby linking glycolysis to the citric acid cycle and ultimately energy production in the cell. PDHX is localized to the mitochondrial matrix. Defects in PDHX cause pyruvate dehydrogenase deficiency which results in lactic acid acidosis and can lead to neurological dysfunction and mental retardation. Interestingly, PDHX is linked to the autoimmune disease called primary biliary cirrhosis (PBC). In PBC, cells in the liver get attacked by activated T cells because of autoantibodies to PDHX, which can ultimately lead to liver disease and liver failure. EMD-Millipore’s Anti-PDHX rabbit polyclonal antibody has been tested in western blot on BxPC-3 cell lysates and in paraffin embedded immunohistochemistry on human colon cancer tissue sections.
