背景信息 Transcription factor 7-like 1, also known as HMG box transcription factor 3 (TCF-3), and encoded by the gene TCF7L1 or TCF3, is a transcription factor that participates in the Wnt signaling pathway and is necessary for the terminal differentiation of epidermal cells, the formation of the keratophyalin granules, and the barrier function of the skin. TCF-3 is localized in the nucleus and expressed in hair follicles and skin keratinocytes and in intestinal epithelium of the stomach and small intestine. TCF-3 is also highly expressed in poorly differentiated human breast cancers, preferentially of the basal-like subtype. This suggests that TCF-3 is involved in the regulation of breast cancer cell differentiation state and tumorigenicity. Silencing of TCF3 dramatically decreased the ability of breast cancer cells to initiate tumor formation, and led to decreased tumor growth rates. In culture, TCF-3 promotes the sphere formation capacity of breast cancer cells and their self-renewal.
