背景信息	Peptidyl-prolyl cis-trans isomerase B, PPIase B, (PPIB) or alternatively CYP-S1, Cyclophilin B, Peptidyl-prolyl isomerase B, Rotamase B, or S-cyclophilin (CYPB) is an endoplasmic reticulum localized protein that assists in the proper folding and coiling of proteins during synthesis. PPIB binds and is inhibited by the immune suppressant drug cyclosporine A. PPIB can also be secreted and when secreted it appears to mediate T cell chemotaxis and adhesion. The PPIB receptor is CD14, and PPIB is a chemotactic factor for bone marrow-derived mesenchymal stromal cells and neutrophils. Overexpression of PPIB suppresses apoptotic cell death induced by reactive oxygen species and Ca++ homeostasis; however overexpression of PPIB is also associated with malignant progression and appears to play some role in breast cancer pathogenesis. Finally, PPIB while chemotactic for macrophages does not induce their induction of inflammatory cytokines such as TNF alpha. On the contrary it seems that PPIB works to attenuate the reactions of proinflammatory macrophages by inducing the expression of Bcl-3 which ultimately decreases the expression of TNF alpha and other associated genes, thus it is now believed that cyclophilin A and cyclophilin B work together to mediate and coordinate as least some proinflammatory responses in macrophages and other immune cells. EMD-Millipore’s Anti-PPIB rabbit polyclonal antibody has been tested in western blot on Raji cell lysates and in paraffin embedded immunohistochemistry on human lymphoma tissues sections.
