The PureGenome™ On-Spot Tissue DNA kit allows for convenient isolation of genomic DNA in any targeted microscopic tissue area on a slide. The system combines a powerful area-specific tissue sampling technology with a one-step DNA extraction method and makes targeted tissue genomic DNA isolation easy and simple. The kit efficiently recovers genomic DNA from paraffin archives or fresh tissue sections for PCR analysis. Unlike a conventional UV-fractionation method, this system does not destroy the surroundings of target cells. There is no need for expensive specialized equipment or computer software. DNA purified using the PureGenome On-Spot Tissue DNA kit is suitable for challenging downstream applications such as Next Generation Sequencing workflows and quantitative PCR.

This kit can be used with fresh tissue sections or paraffin archives. It can also be used with frozen sections and tissue sections fixed by solvents such as acetone, ethanol, methanol, etc.

Method Principle: As outlined in Figure 1(bottom of page), simply apply the Spot Solution to a selected microscopic tissue area. The solution will air-dry into a thin film while the underlying cells will become embedded within. The area is lifted and transferred into a tube. After digestion with the one-step Extraction Buffer (containing proteinase K) the DNA is ready for PCR analysis. Figure 2 (bottom of page) illustrates a representative lot of PCR amplified DNA fragments from different samples of paraffin embedded tissue.

One kit is sufficient for 50 test samples.

Storage: Upon initial preparation, add 260 µl of Proteinase K Storage Buffer to the Proteinase K tube. Dissolve completely and then store at –20°C. Store all other components at room temperature. This kit is stable for at least one year at the recommended storage temperature. Reagents are routinely tested on lot-to-lot basis to ensure they provide maximal performance and reliability.
