概述	All ChIPAb+ antibodies are individually validated for chromatin precipitation, every lot, every time. Each ChIPAb+ antibody set includes control primers (tested every lot by qPCR) to biologically validate your IP results in a locus-specific context. The qPCR protocol and primer sequences are provided, allowing researchers to validate ChIP protocols when using our antibody in their chromatin context. Each set also includes a negative control antibody to ensure specificity of the ChIP reaction. The ChIPAb+ SOX-2 set includes the SOX-2 antibody, a Normal mouse IgG, and control primers which amplify a 95 bp region of ChIP Primers, nanog promoter. The SOX-2 and negative controls are supplied in a scalable "per ChIP" reaction size and can be used to functionally validate the precipitation of SOX-2 -associated chromatin.

背景信息	SOX 2 is also known as SRY related HMG BOX gene 2. All SOX proteins have a single HMG box and bind linear DNA in a sequence-specific manner, resulting in the bending of DNA through large angles. Bending causes the DNA helix to open for some distance, which may affect binding and interactions of other transcription factors. SOX1, SOX2 and SOX3 show the closest homology to SRY. They share maximum homology within the HMG domain and are expressed mainly in the developing nervous system of the mouse. These genes share significant homology outside the HMG box also and are highly conserved throughout their evolution. Sox-2 plays a key role in embryonic stem cell development, and has been shown to help stem cells maintain a pluripotent potential. It has also been shown to be associated with uncommitted dividing stem cell and precursor cells of the developing CNS.

应用说明	Chromatin Immunoprecipitation: Representative lot data. Sonicated chromatin prepared from NCCIT cells (~2 X 10E6 cell equivalents per IP) were subjected to chromatin immunoprecipitation using 3 µg of either Normal mouse IgG or 3 µg of Anti-SOX-2 and the Magna ChIP™ G Kit (Cat. # 17-611). Successful immunoprecipitation of SOX-2 associated DNA fragments was verified by qPCR using ChIP Primers, nanog promoter as a positive locus, and MyoD promoter primers as a negative locus. (Figure 2). Data is presented as percent input of each IP sample relative to input chromatin for each amplicon and ChIP sample as indicated. Please refer to the EZ-Magna ChIP™ G (Cat. # 17-409) or EZ-ChIP™ (Cat. # 17-371) protocol for experimental details. Western Blot Analysis: Representative lot data. Mouse embryonic stem cell lysate was probed with Anti-SOX-2 (0.5 μg/mL). Proteins were visualized using a Goat Anti-Mouse IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates SOX-2 (~34 kDa). (Figure 3). Immunocytochemistry Analysis: Representative lot data. RenCX cells were stained with 10 μg/mL of Cat. No. MAB4423, Anti-SOX-2. Reactivity was detected using a Goat Anti-Mouse secondary antibody in Hoescht stain. Positive staining was observed in RenCX cells.
