背景信息	Induced pluripotent stem (iPS) cells can be generated from somatic cells by enforced expression of four Yamanaka transcription factors, Oct4, Klf4, Sox2, and c-Myc (Takahashi and Yamanaka, 2006). True pluripotency is defined as the ability to give rise to all cell types except for extra-embryonic tissues. Recent findings suggest that factor-induced reprogramming primarily generates an undifferentiated intermediate cell state that is not completely pluripotent (Jaenisch and Young, 2008). These partially reprogrammed cells (pre-iPS or Class I iPS cells) have global gene expression and DNA methylation patterns that are distinct from ES cells in spite of similaries in colony morphology and the ability to be propagated extensively in culture. Partial reprogrammed cells are characterized by the continued expression of viral transgenes, the incomplete expression of pluripotent genes such as Nanog and Rex1, down-regulation of somatic cell marker genes, and the inability to generate adult chimeric mice and transmit through the germ line (Silva J, 2008). The STEMCCA Viral Gene Detection qPCR Multiplex Kit (Human) will help determine whether an iPS clone generated with the STEMCCA lentivirus is a fully reprogrammed colony or a partially reprogrammed intermediate. This real time PCR kit has been designed to detect STEMCCA viral transgene expression along with the upregulation of human Nanog, a key ES/iPS cell pluripotency marker, in a multiplex setting enabled by the proprietary Amplifluor® technology.
