概述	MilliTrace™ CX Nestin GFP Reporter Human Neural Stem Cell Kit (Catalog No. SCR096) provides ready-to-use human neural stem cells that are labeled with the humanized mulleri green fluorescent protein (hmGFP) under the regulation of the Nestin promoter along with expansion medium to help maintain expression of the transgene. The parental cell line, ReNcell CX, is an immortalized human neural progenitor cell line with the ability to readily differentiate into neurons and glial cells. The ReNcell CX line was derived from the cortical region of a human fetal brain. Immortalized by retroviral transduction with the myc oncogene, this cell line grows rapidly as a monolayer on laminin with a doubling time of 20-30 hours. MilliTrace CX Nestin GFP Reporter Human Neural Stem Cells were generated by electroporation of the ReNcell CX line with a proprietary bicistronic plasmid containing hmGFP under the control of the mouse Nestin promoter. Nestin is an intermediate filament protein expressed in proliferating neural progenitors and stem cells during the early stages of development in the CNS and PNS. Expression of Nestin is commonly associated with undifferentiated neural stem and early progenitor cells. Upon differentiation, Nestin is down-regulated and since hmGFP is under the regulation of the mouse Nestin promoter, GFP expression correspondingly declines upon further differentiation. FACS analyses of stable proliferating transfectants indicate that greater than 95% of the cells express GFP at high levels even after 10 passages. MilliTrace CX Nestin GFP Reporter Human Neural Stem Cells display the immunochemical staining properties of neural stem cells (NSCs); they are positive for Nestin and Sox-2 expression. The cells are multipotent and are able to differentiate into neurons and astrocytes. Upon differentiation, GFP expression is significantly downregulated. Cells have been confirmed to be mycoplasma-free and demonstrate an apparently normal karyotype (46, XY) as assessed by standard G-banding analysis performed on 20 metaphase cells.

Upon thawing the basal medium should be stored at 2-8°C and given a 1-month expiration dating. 5 mg/mL Puromycin Solution, 100 μL (Part No. CS201361) should be stored in working aliquots at -20°C for up to 2 years.
