背景信息 Aplysia Ras-related homologs (ARHs) are another name for Rho proteins. Rho proteins belong to a superfamily of small guanine nucleotide binding/exchange proteins vital to many intracellular signaling cascades. These small guanine binding proteins can be held in an inactive state by binding particular intracellular inhibitors. Rho GDP-dissociation inhibitor 1 (Rho GDI 1) or alternatively Rho-GDI alpha, encoded by the gene named ARHGDIA or GDIA1 is one such inhibitor. ARHGDIA is a small cytoplasmic protein that can keep GTP from binding Rho proteins by keeping GDP from disassociating from them. By mediating such a reaction, ARHGDIA mediates the signaling cascades involving RHOA, CDC42, RHOH, RAC1 and RAC2. Rho-GTPases are critical regulators of many cellular activities and have been termed "molecular switches" that play critical roles in cell proliferation, apoptosis, and multiple gene expression pathways. ARHGDIA, through the control of Rho proteins, also plays an important role in control the intracellular dynamics of actin and the functioning of the cytoskeleton, which in turn controls such things as cell motility and migration and even cellular functions such as in podocytes and the proper operation of the glomerular filtration barrier. Defects in ARHGDIA can lead to inappropriate activation of cellular processes which can lead to Ras/Rho induced tumorigenesis, and recent research may indicate that ARHGDIA may be useful as a diagnostic biomarker and/or a therapeutic to prevent colon and prostate cancer metastasis. EMD-Millipore’s Anti-ARHGDIA mouse monoclonal antibody has been tested in western blot on recombinant protein as well as cell lysates from Jurkat, HeLa, NIH3T3, C6, K562 and COS7 cells and fluorescent immunocytochemistry on HepG2 cells in culture.
