背景信息	The protein Serine/threonine-protein kinase WNK1 (WNK1), or alternatively known as Erythrocyte 65 kDa protein (p65), Kinase deficient protein, Protein kinase lysine-deficient 1, and Protein kinase with no lysine 1 (hWNK1), and encoded by the human gene named WNK1 or HSN2, KDP, KIAA0344, PRKWNK1 is a serine/threonine kinase that plays an critical role in the regulation of electrolyte homeostasis, signal transduction, cell survival and cell proliferation. WNK1 functions to activate or deactivate various ion channel transporters including sodium-coupled chloride cotransporters and potassium coupled chloride cotransporters including SCNN1A, SCNN1B, SCNN1D and SGK1. Additionally WNK1, via its kinase activity, appears to play a role in cytoskeletal reorganization and even apoptosis regulation. WNK1 typically associates with its inhibitor partner WNK4 and both WNK1 and WNK4 levels are controlled at least partial via ubiquitination. WNK1 also has different isoforms due to what seems to be largely tissue specific splicing events and the isoforms appear to be regulated developmentally as well, particularly in the developing nervous and sensory tissues. Finally, WNK1 knockouts have been shown to be an embryonic lethal in endothelial-specific gene knockouts and WNK1 plays an essential role in regulating angiogenesis and cardiac formation during development. EMD-Millipore’s Anti-WNK1 rabbit monoclonal antibody has been tested in western blot on Saos-2 cell lysates as well as tissue lysates from fetal heart and kidney. The antibody has also been successfully used in paraffin embedded immunohistochemistry on normal human kidney tissue sections.
