背景信息	Meningioma-expressed antigen 5 (MGEA5), also known as bifunctional protein NCOAT, or Nuclear cytoplasmic O-GlcNAcase and acetyltransferase, with the gene names MGEA5, HEXC, KIAA0679, and MEA5 is a bi-functional enzyme, in other words, an enzyme that contains kinds of activity. For MGEA5 it has a O-GlcNAcase domain as mentioned above, (this activity also has alternative names including Glycoside hydrolase O-GlcNAcase, Hexosaminidase C, N-acetyl-beta-D-glucosaminidase, N-acetyl-beta-glucosaminidase, O-GlcNAcase (OGA)) and a histone acetyltransferase (HAT) domain. MGEA5 can cleave GlcNAc but not GalNAc bonds in glycopeptides and acetylates lysine -8 (Lys-8) of histone H4 and lysine-14 of histone H3. MGEA5 O-GlcNAcase active site lies in the N terminus of the enzyme and its histone acetyltransferase (HAT) domain lies in the C terminus. The HAT domain MGEA5 resembles that of other acetyltransferases, but MGEA5 also contains a motif resembling a zinc finger-like domain that is unique to the MYST family of HATs. The zinc finger-like domain is responsible for making contacts with the histone tails within nucleosome allowing the acetylation to occur. The protein demonstrates the highest level of expression in the brain, placenta and pancreas during development and adult life; MGEA5 is proteolytically cleaved by caspases-3 during apoptosis activations as well. EMD-Millipore’s Anti-MGEA5 rabbit monoclonal antibody has been tested in western blots on human cell lysates from HeLa, BxPc-3 and JAR cell lines as well as human fetal brain lysates. The antibody has also been tested in immunohistochemistry on paraffin embedded human kidney tissue sections.
