背景信息	CD73 is now known to be an enzyme called 5'-nucleotidase, or ecto-5-prime-nucleotidase with a gene name of NT5E, NT5, or NTE, and is the protein located on the plasma membrane. CD73 hydrolyzes extracellular nucleotides into membrane permeable nucleosides and free phosphate. Its preferred substrate is AMP, and CD73 is a key enzyme in the regulation of adenosinergic signaling. The enzyme consists of a dimer of 2 identical 70-kD subunits bound by a glycosyl phosphatidyl inositol linkage to the external face of the plasma membrane. In immunology, CD73 is a marker for mature lymphocytes. Consequently, a deficiency of CD73 occurs in a variety of immunodeficiency diseases. Mammalian cells contain various other isozymes with similar activities and they are distinguished from CD73 by their substrate affinities, requirement for divalent magnesium ion, activation by ATP, and inhibition by inorganic phosphate. Interestingly, recent research indicates that inhibiting CD73 activity could help treat cancer and prevent metastasis, and that CD73 is necessary for effective T-lymphocyte invasion in the brain under various immunological disease states, so by inhibiting its activity neuroinflammation could be potentially be mediated. EMD-Millipore’s Anti-CD73 rabbit monoclonal antibody has been tested in western blot on human fetal liver and adult spleen tissue lysates as well as human A375 cell lysates. The antibody has also been tested in immunohistochemistry on paraffin embedded human tonsil tissue sections.
