背景信息	Zinc finger protein GLI1 (Gli-1), or Glioma-associated oncogene is a transcriptional activator that becomes mutated and activated in certain gliomas but whose normal role appears to be in mediating sonic hedgehog signaling (SHH) in craniofacial development and digital development, as well as in the development of the central nervous system and gastrointestinal tract. Because Gli-1 is a mediator of SHH, Gli-1 is important in the adult as well because SHH controls cell division of adult stem cells and SHH has been implicated in the development of certain cancers. The protein shuttles between the cytoplasm and nuclear locations; Gli-1 is tethered in the cytoplasm by binding the protein SUFU, another protein in the SHH pathway. Activation and translocation to the nucleus is promoted by interaction with STK36 (Serine/threonine-protein kinase 36) where Gli-1 will interact with another zinc-finger transcription factor partner ZIC1 to enhance gene transcription. Expression of Gli-1 is normally developmentally regulated and can be seen in testis, myometrium and fallopian tube. Gli-1 is also expressed in the brain with highest expression in the cerebellum, optic nerve and olfactory tract. EMD-Millipore’s Anti-Gli-1 rabbit monoclonal antibody has been tested in western blot on human cell line lysates as well as in immunohistochemistry on paraffin embedded human brain tissue sections.
