背景信息	STIP1 (Stress-induced-phosphoprotein 1), STI1, also known as Hsp70-Hsp90 Organizing Protein (Hop, HOP), Renal carcinoma antigen NY-REN-11, and Transformation-sensitive protein IEF SSp 3521, belongs to a large group of proteins known as co-chaperones which help to regulate the actions of the major chaperones, which are mainly heat-shock proteins. The STIP1 family of proteins is a large family of co-chaperones that is evolutionarily conserved being found from viruses and parasites to plants, fungi and mankind. The family is identified by having a unique nine tetratricopeptide repeat (TPR) that are clustered into domains of three TPRs. It is through the interaction and binding of the various TPR repeats that the STIP1 family members function. STIP1 functions as an adaptor protein that coordinates the functions of heat-shock protein 70 (HSP70) and heat-shock protein 90 (HSP90) in protein folding. STIP1 facilitates the transfer of proteins from HSP70 to HSP90 by binding both HSP90 and substrate-bound HSP70. STIP1 also stimulates the ATPase activity of HSP70 and inhibits the ATPase activity of HSP90, suggesting that STIP1 regulates both the conformations and ATPase cycles of these heat-shock proteins. STIP1 localizes to both the nucleus and cytoplasm depending upon the conditions. Interestingly STIP1 participates in other non-chaperone protein complexes too such as with Hepatitis B virus and its reverse transcriptase complex thereby allowing viral replication, and STIP1 acts as a cellular receptor for prion proteins. EMD-Millipore’s Anti-STIP1 rabbit monoclonal antibody has been tested in western blot on various human cancer cell lines and in immunohistochemistry on paraffin embedded human ovarian cancer tissue sections.
