背景信息	Dynamin is a 100 kDa GTP binding protein that was first identified as a protein able to bind microtubules. A mutation in the Drosophila gene shits, encoding a dynamin homologue, prevents synaptic vesicle recycling and causes temperature-sensitive paralysis, implicating dynamin in the formation of coated vesicles. Dynamin may mediate vesicle formation by assembling into a ring structure around the necks of invaginated coated pits that may eventually lead to the closure of the neck and formation of vesicles. Amphiphysin, dynamin, clathrin and AP2 form a complex during synaptic vesicle endocytosis, which is regulated by the phosphorylated state of different components in this complex. To date, three genes have been found to encode dynamin isoforms. Dynamin 1 is neuronal specific, Dynamin 2 is ubiquitously expressed and Dynamin 3 localizes to testis, brain and lung.

应用说明	Western blot on human, rat and mouse brain homogenate: Brains were isolated rodents and homogenized at 2,800 rpm with a Potter-Elvehjem type homogenizer after adding 3 vol of 4°C Buffer A (10 mM Hepes, pH 7.2, 25 mM sucrose, and 10 mM EGTA) containing protease inhibitors (1 mM phenylmethylsulfonyl fluoride, 100 μg/ml soybean trypsin inhibitor, 10 μg/ml leupeptin, 10 μg/ml p-toluenesulfonyl-L-arginine methyl ester, and 5 μg/ml pepstatin A). The homogenate was centrifuged 15 min at 13,000 g then 1 h at 100,000 g, and the supernatant was collected as crude synaptosomal fraction. This fraction was subjected to SDS-PAGE and Western blot analysis. Detects a band of approximately 100 kDa (97.4kDa). Immunofluorescence on rat brain tissue that has been perfused and fixed with formaldehyde, 7 micron cut sections frozen sections. Has not been tested on paraffin embedded tissue sections. Immunoprecipitation on Triton X-100 extracts of post-nuclear supernatant of rat brain homogenate: brains from male Sprague Dawley rats were homogenized as described above in 4 vol of 4°C RIPA buffer (50 mM Tris, pH 8.0, 150 mM NaCl, 1 mM EDTA, 1% Triton X-100, 0.5% deoxycholate, and 0.1% SDS) containing protease inhibitors. The homogenate was centrifuged as above and the supernatant collected as the clarified extract. Appropriate antibodies (5 μg) were incubated with 1-ml fractions (15-25 mg) of the clarified extracts, and then preswollen protein G beads (5 mg) were added before a second incubation. Incubations were 2-12 h at 4°C with rotation. Beads and immune complexes were collected by centrifuging 3 min at 7,000 g and then rinsed 4 × 5 min with the starting buffer before being subjected to SDS-PAGE and Western blot analysis. Optimal working dilutions must be determined by end user.
